The microtiter plate provided in ELISA Kit has been precoated with an antibody specific to the target protein. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated specific polyclonal antibody preparation,and Avidincon jugated to Horseradish Peroxidase（HRP） is added to each microplate well and incubated. Then a TMB substrate solution is added to each well. Only those wells that contain target protein, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm ± 2 nm. The concentration of the samples is then determined by comparing the O.D. of the samples to the standard curve.

自备物品
1. 酶标仪（建议参考仪器使用说明提前预热）
2. 微量加液器及吸头，EP 管
3. 蒸馏水或去离子水，全新滤纸 标本的采集及保存
1. 血清：全血标本请于室温放置 2 小时或 4℃过夜后于 1000 x g 离心 20 分钟，取上清即可
检测，或将标本放于-20℃或-80℃保存，但应避免反复冻融。
2. 血浆：可用 EDTA 或肝素作为抗凝剂，标本采集后 30 分钟内于 2 - 8° C 1000 x g 离心 15
分钟，或将标本放于-20℃或-80℃保存，但应避免反复冻融。
3. 细胞培养物上清或其它生物标本：请 1000 x g 离心 20 分钟，取上清即可检测，或将标本
放于-20℃或-80℃保存，但应避免反复冻融。
注：以上标本置 4℃保存应小于 1 周，-20℃或-80℃均应密封保存，-20℃不应超过 1 个月，-80℃
不应超过 2 个月；标本溶血会影响zui后检测结果，因此溶血标本不宜进行此项检测。
样本处理：血清或血浆标本*稀释 10 倍，如：稀释 10 倍，取 100uL 血清或血浆加入 900uL
样品稀释液。建议各实验室在操作前*行预实验以建立*稀释倍数。
