背景介绍：The protein encoded by this gene was first identified by its binding to the APC protein which is often mutated in familial and sporadic forms of colorectal cancer. This protein localizes to microtubules, especially the growing ends, in interphase cells. During mitosis, the protein is associated with the centrosomes and spindle microtubules. The protein also associates with components of the dynactin complex and the intermediate chain of cytoplasmic dynein. Because of these associations, it is thought that this protein is involved in the regulation of microtubule structures and chromosome stability. This gene is a member of the RP/EB family.

关于肽链的设计

多肽是大分子，每条多肽序列在物理和化学特性上都有自己的*性。有些多肽合成起来很困难，还有些合成相对容易，但纯化困难，其主要是不溶于水，所以在纯化中，那些疏水肽必须溶于非水溶剂中或特殊的缓冲液，然而这些溶剂或缓冲液可能不适合应用于生物实验系统，研究人员不能使用该多肽达到研究目的。

对研究人员的多肽设计提供一些建议：

一、变难为易

1、减少序列长度

肽的长度增加导致粗产物纯度降低，小于15个残基的肽较容易得到.当肽链增加到20个以上时，正常产物的量就要考虑。在具体实践中残基低于20往往能得到更好的结果；

2、减少疏水残基

疏水残基占明显优势的肽，尤其在距C端7―12个残基的区域，会引起合成困难，一般认为由于合成中形成β折叠片，而产生不*配对。当然，可用几个极性残基置换，或加入Gly或Pro易打开肽结构可能会得到帮助；

3、减少“难度”残基

有多个Cys、Met、Arg、Try残基一般难以合成，Ser通常可作为非氧化替换。
